This is yet another irksome quirk with Worldbuilder. If you leave all starting locations as "Random Player", and allow the player to select from any of the major civilizations you have assigned as being usable within the 'scenario', what will happen is that the human player will always start at one of the pre-assigned starting points, and irregardless of who you select to play as or how many times you start a game using the precreated map, you will always start in that same spot. You can also assign specific plots as starting positions for city-states, and for them I generally assign them as Random City-State start positions. Once you fill the list of civs you want as part of the map set-up, you can assign individual tiles as either Edit Plots Tab > "Start Position" = Random Player or you can specify which player will start in on which tile.
I think but have never tested that you will get insta-lose if you select only one player for the map, but choose to use a different player when starting with a premade map. If you select only 1 player, and you play as that player using the map, you will get insta-win as soon as you start the game. The default usage will be for the human player to be Player 1.
